Estudio cuantitativo y composicional sobre biofilms monoespecie de microorganismos alterantes para la industria cárnica e interacción en el desarrollo de biofilms multiespecie

Resum: La alteración de los alimentos constituye un grave problema económico para la industria alimentaria, generando pérdidas importantes. Un alimento alterado no necesariamente tiene que suponer un riesgo para la salud pública, pero carece de la calidad que los consumidores esperan de él. Una de las principales causas de estas alteraciones es la contaminación con microorganismos que persisten en la industria en forma de biofilms, lo cual dificulta su control. En el estudio que se presenta a continuación, se utilizaron tres especies distintas de microorganismos alterantes: Pseudomonas fragi, Leuconostoc gasicomitatum y Lactobacillus reuteri, todos ellos presentes en la carne y tracto gastrointestinal del pollo. El estudio consistió en determinar su capacidad de formación de biofilms monoespecie así como la interacción que tienen cuando éstos se encuentran en convivencia. De este modo, se utilizaron las tres especies para investigar la formación de biofilms simples maduros en una semana a una temperatura de 30ºC. L. gasicomitatum se utilizó también para investigar el comportamiento de P. fragi y L. reuteri en la formación de biofilms multiespecie sobre superficies de acero inoxidable. La evaluación de la estructura y composición de los biofilms monoespecie se realizó mediante microscopía de epifluorescencia directa (DEM). En el caso de los biofilms multiespecie, el crecimiento se determinó por recuento de colonias en placa. Los resultados demostraron que tanto L. gasicomitatum como L. reuteri fueron capaces de formar biofilms, obteniendo recuentos aproximadamente de 7 Log UFC/cm2 y estructuras definidas. Contrariamente, P. fragi obtuvo recuentos del orden de 4 Log UFC/cm2, significativamente diferentes a las especies anteriores (P < 0. 05), y no presentó una red de conglomerados celulares típico en biofilms. En relación a las macromoléculas que conformaron la matriz, se obtuvo entre 70 - 80 % de contenido proteico para los biofilms de L. gasicomitatum y L. reuteri, a diferencia de P. fragi que presentó un mayor contenido polisacárido (P < 0. 05). Los resultados de los biofilms multiespecie demostraron que la presencia de P. fragi no afectó al crecimiento de L. gasicomitatum, el cual mantiene su crecimiento de entre 5,76 a 6,1 Log UFC/cm2. Sin embargo, L. reuteri podría ejercer un desplazamiento de L. gasicomitatum tras 24 horas en convivencia como biofilm mixto, al presentar una diferencia en el recuento de aproximadamente 2 Log UFC/cm2. El estudio de biofilms, tanto monoespecie como multiespecie, son de interés pues ayuda comprender las relaciones en ecología microbiana y a buscar nuevas tecnologías para su eliminación.
Resum: The alteration of food represent a serious economic problem in food industry, creating significant losses. An altered food does not necessarily have to pose a risk to public health, but it lacks the quality that consumers expect from it. One of the main causes of these alterations is the contamination with microorganisms that persist in the industry in the form of biofilms, which makes their control difficult. In the study presented below, three different species of alternating microorganisms were used: Pseudomonas fragi, Leuconostoc gasicomitatum and Lactobacillus reuteri, which can be found in the meat and gastrointestinal tract of the chicken. The main aim of this study was to determine their ability to form monospecies biofilms as well as the interaction they have when they coexist with one another. Thus, the three species were used to investigate the formation of mature simple biofilms in a week at a temperature of 30 ° C. L. gasicomitatum was also used to investigate the behavior of P. fragi and L. reuteri in the formation of multispecies biofilms on stainless steel surfaces. The evaluation of the structure and composition of the monospecies biofilms was performed by direct epifluorescence microscopy (DEM). In the case of multispecies biofilms, growth was determined by plate colony count. The results showed that both L. gasicomitatum and L. reuteri were able to form biofilms, obtaining counts of approximately 7 Log CFU / cm2 and defined structures. On the contrary, P. fragi obtained counts of the order of 4 Log CFU / cm2, significantly different from the previous species (P <0. 05), and did not present a network of typical cell clusters in biofilms. In relation to the macromolecules that formed the matrix, between 70-80% protein content was obtained for the biofilms of L. gasicomitatum and L. reuteri, unlike P. fragi that presented a higher polysaccharide content (P <0. 05). The results of the multiespecies biofilms showed that the presence of P. fragi did not affect the growth of L. gasicomitatum, which maintains its growth between 5. 76 to 6. 1 Log CFU / cm2. However, L. reuteri could exert a displacement of L. gasicomitatum after 24 hours in coexistence as a mixed biofilm, presenting a difference in the count of approximately 2 Log CFU / cm2. The study of biofilms, both monospecies and multispecies, are of interest because it helps to understand the relationships in microbial ecology and to look for new technologies for their elimination.
